Abstract
Uvod: Visceralna bol je bol koja proizlazi iz oštećenja visceralnih organa. Prenosi se nemijeliziranim, C-vlaknima. Uzrokuje veliko smanjenje kvalitete života u bolesnika koji pate od autoimunih demijelinizirajućih bolesti. Pomoću pokusnih modela demijelinizacije, miševa hranjenih kuprizonom i miševa sa isključenim genom B4Galnt1, moguće je pratiti pojavu i molekularnu podlogu fenotipa visceralne boli. Ciljevi: Markerima mijelinizacije, maturacije i neurodegeneracije aksona procijeniti oštećenje aksona somatosenzoričkih putova. Uz pomoć epitopa NeuN i nNOS kvantificirati neurone u ventralnom, lateralnom i dorzalnom rogu kralježnične moždine lumbalnog segmenta na sve tri skupine životinja. Markerima upale, Iba1 i GFAP, procijeniti stupanj upale u putovima za bol. Materijali i metode: U istraživanje su uključena tri tipa miševa - genetički modificirani B4Galnt1 (KO), tretirani kuprizonom (CUP) i miševi divljeg tipa (WT). Imunohistokemijskom analizom pomoću protutijela MBP, CNP-aza, Iba1, GFAP, SMI312, pTau, NeuN i nNOS detektirani su pripadaući biljezi. Rezultati: KO i CUP skupina imaju nižu ekspresiju svih markera mijelinizacije, upale, neurodegeneracije i neurofilamenata u odnosu na WT skupinu. Niti u jednoj skupini nema umiranja neurona uslijed demijelinizacije. Zaključak: Procesi demijelinizacije i upale dovode do demijelinizacije somatosenzoričkih vlakana i pojačanja fenotipa visceralne boli.
Abstract (english)
Introduction: Visceral pain is the one that derives from visceral organs. It is conducted by unmyelinated, C-fibers. Visceral pain causes degradation in quality of life in patients who suffer from autoimmune demyelinating diseases. With the help of two demyelination models, mice that are fed with cuprizone and the one with knock-out gene, B4Galnt1, it is possible to follow perception and molecular basis of sensitivity to visceral pain. Goals: With the help of markers of myelination, maturation and degeneration in axons estimate the damage in axons of somatosensory pathways. With epitopes NeuN and nNOS quantify neurons in ventral, lateral and dorsal horn in lumbar part of the spinal cord in all three groups of animals. With the markers of inflammation, Iba1 and GFAP, determine level of inflammation in the pain pathways. Materials and methods: In this research were included three groups of animals – genetically modified B4Galnt1 knock-out, group treated with cuprizone and wild type group. Immunohistochemical analysis with antibodies MBP, CNPase, Iba1, GFAP, SMI312, pTau, NeuN and nNOS was used to detect belonging epitops of interest. Results: Collected results show lower expression of all markers in comparison with WT group, except neuron markers, which in this group of animals do not die. Conclusion: Process of demyelination causes demyelination of somatosensory neurons and increased perception of viceral pain.
