Abstract
CILJEVI ISTRAŽIVANJA: Cilj ovog istraživanja uspoređivanje je različitih morfoloških karakteristika bubrežnog tkiva između miševa divljeg tipa i miševa s isključenim genom Tff3 kojima je induciran stres endoplazmatske mrežice. USTROJ STUDIJE: Istraživanje je provedeno kao presječna studija u kojoj su se koristili uzorci mišjih bubrega iz arhiva Zavoda za histologiju i embriologiju. MATERIJAL I METODE: Za istraživanje su korišteni histološki preparati bubrega miševa divljeg tipa i onih s isključenim genom Tff3 kojima je tunikamicinom induciran stres endoplazmatske mrežice 24h prije žrtvovanja. Preparati su bojani PAS metodom. Korišteno je 5 miševa iz svake skupine i načinjeno 5 fotografija kore i 3 fotografije srži za svaku životinju na kojoj su mjerene vrijednosti površine presjeka glomerula, promjera kanalića, visine epitela kanalića i promjera lumena kanalića. REZULTATI: Nije bilo statistički značajne razlike u površini presjeka glomerula, promjeru kanalića niti u visini epitela i promjeru lumena kanalića miševa divljeg tipa i miševa s isključenim genom Tff3 kojima je induciran stres endoplazmatske mrežice. ZAKLJUČAK: Ne postoje značajne razlike u ispitivanim morfološkim obilježjima tkiva bubrega u miševa divljeg tipa i miševa s isključenim genom Tff3 kojima je induciran stres endoplazmatske mrežice. Nedostatak Tff3 proteina ne utječe značajno na odgovor stanica bubrega na stres endoplazmatske mrežice.
Abstract (english)
OBJECTIVES: The objective of this study was to compare different morphological characteristics of kidney tissue between wild-type mice and Tff3 knock-out mice with induced endoplasmic reticulum stress. STUDY DESIGN: This research was done as a cross-sectional study using mouse kidney samples from the archives of the Department for Histology and Embryology. MATERIAL AND METHODS: Histological samples of the kidneys of wild-type mice and Tff3 knock-out mice with tunicamycin induced endoplasmic reticulum stress 24 h before sacrifice were used for this study. The samples were stained with the PAS method. 5 mice from each group were used for the study and 5 pictures of the medulla and 3 pictures of the cortex for each animal were taken on which values of glomerular area, tubular diameter, tubular cell height and tubular lumen diameter were measured. RESULTS: There weren't any significant differences in the glomerular area, tubular diameter, tubular cell height and tubular lumen diameter between wild-type mice and Tff3 knock-out mice with induced endoplasmic reticulum stress. CONCLUSION: There are no significant differences in examined morphological characteristics of kidney tissue between wild-type mice and Tff3 knock-out mice with induced endoplasmic reticulum stress. Tff3 protein deficiency does not affectt cellular mechanisms responsible for changes after endoplasmic reticulum stress.
