Abstract
CILJEVI ISTRAŽIVANJA: Glavni je cilj ovog istraživanja otkriti razliku metabolita eritrocita u ljudi sa slezenom i onih bez slezene putem slikovne masene spektrometrije. Drugi cilj je izmjeriti hematološke i biokemijske parametre iz krvi jedne i druge skupine te na temelju njih uočiti moguće razlike. NACRT STUDIJE: Istraživanje je provedeno kao studija parova. ISPITANICI I METODE: Eksperimenalnu skupinu sačinjava 5 osoba koje su bile podvrgnute splenektomiji dok kontrolnu skupinu čini 5 osoba koje odgovaraju po spolu i dobi osobama iz eksperimentalne grupe. Ispitanicima su izmjereni hematološki i biokemijski parametri iz krvi radi uočavanja značajne razlike između grupa te je od njihovih uzoraka napravljen krvni razmaz kojega smo analizirali MALDI TOF uređajem. Za usporedbu laboratorijskih pretraga iz krvi dviju nezavisnih grupa koristišten je t-test. Prilikom obrade uzeta je razina rizika od 5 %. Za obradu podataka korišten je računalni program za statističku obradu IBM SPSS (inačica verzija 24.0, SPSS Inc., Chicago) te ImageReveal (Shimadzu, Kyoto, Japan). REZULTATI: Prema rezultatima laboratorijskih pretraga iz krvi, splenektomirane osobe, prema našem istraživanju, u prosjeku pokazuju višu razinu leukocita, glukoze, slobodnih lakih lanaca kapa i lambda te beta globulina od osoba koje imaju slezenu. MSI analiza krvnih razmaza pokazuje višu razinu ATP-a u zdravih kontrola. ZAKLJUČAK: Splenektomija dovodi do oscilacije koncentracija imunoloških parametara mjerenih iz krvi. Splenektomija bi mogla kao posljedicu imati povećane razine glukoze u krvi. Eritrociti splenektomiranih osoba bi mogli imati nižu razinu ATP-a.
Abstract (english)
OBJECTIVES: The main goal of the research was to discover the differences in the red blood cells metabolites in people with a spleen versus people without the spleen via imaging mass spectrometry. Other goals were to measure and differentiate the bloods hematological and biochemical parameters in both groups. STUDY DESIGN: Research was conducted as match pair study. SUBJECTS AND METHODS: Experimental group consisted of 5 people who had splenectomy and control group consisted of 5 people who matched the experimental group by age and sex. Blood hematological and biochemical parameters were measured for differentiation and blood smears were analyzed by MALDI TOF device. To compare laboratory tests in both groups t-test was used. A risk level of 5 % was calculated during the tests. For statistical analysis and data processing computer programs IBM SPSS (version 24.0, SPSS Inc., Chicago) and ImageReveal (Shimadzu, Kyoto, Japan) were used. RESULTS: According to laboratory tests of blood samples in conducted research people who had had splenectomy show higher levels of leucocytes, glucose, free kapa and lambda chains and beta globulins compared to people who had not been splenectomised. MSI analysis of blood smears shows higher levels of ATP in healthy individuals. CONCLUSION: Splenectomy does affect oscillation of immunological parameters concentrations measured from blood samples. As a result of splenectomy there is a possibility for higher blood glucose levels and lower erythrocytes ATP levels.
