Abstract
CILJEVI ISTAŽIVANJA. Ciljevi ovoga istraživanja bili su utvrditi postoje li razlike u promjerima jetrenih režnjića miševa, vezivnom tkivu jetre te u prisutnosti glikogena u jetri divljeg tipa i miševa kojima su isključeni TFF2 i TFF3 geni. USTROJ STUDIJE. Preparati jetre analizirani su svjetlosnim mikroskopom kvalitativno, kvantitativno mjerenjem promjera jetrenih režnjića, a semikvantitativno je određena jačina signala glikogena. MATERIJAL I METODE. Istraživanje je provedeno na jetri miševa divljeg tipa i jetre miševa s isključenim genom TFF2 i TFF3. Preparati jetre su bojani trima različitim bojenjima hemalaun i eozinom, PAS bojenjem i Mallory metodom. Izmjereni su promjeri jetrenih režnjića te je brojanjem pozitivnih stanica određena količina glikogena. Kvalitativno je opisana količina vezivnog tkiva u jetrenim režnjićima. REZULTATI. Miševi kojima je isključen TFF2 gen i miševi kojima je isključen TFF3 gen imali su značajno manji promjer jetrenih režnjića nego miševi divljeg tipa. Miševi s isključenim TFF2 i TFF3 genom imali su više vezivnog tkiva u jetrenim režnjićima nego miševi divljeg tipa. Miševi s isključenim TFF3 genom imali su više stanica pozitivnih na glikogen od miševa s isključenim TFF2 genom i miševa divljeg tipa, koji se međusobno nisu statistički razlikovali po količini glikogena. ZAKLJUČAK. Postoje značajne histomorfološke razlike u građi jetre divljeg tipa i miševa kojima je isključen TFF2 i TFF3 gen u promjeru režnjića, količini glikogena u stanicama te vezivnog tkiva u režnjićima.
Abstract (english)
OBJECTIVES: The objectives of this study were to determine if there are differences in the diameter length of hepatic lobules, hepatic connective tissue and the presence of glycogen in the liver of wild type mice and TFF2 and TFF3 knock-out mice. STUDY DESIGN: Liver slides were analyzed under a light microscope qualitatively, quantitatively by measuring the diameter of hepatic lobules, with the glycogen signal strength being determined semi quantitatively. MATERIAL AND METHODS: The study was carried out on liver of wild type mice and TFF2 and TFF3 knock-out mice. The liver slides were stained with three different methods hemalaun and eosin, PAS staining, and Mallory method. The diameters of hepatic lobules were measured, and glycogen quantity was determined by counting the positive cells. The amount of connective tissue in hepatic lobules was described qualitatively. RESULTS: TFF2 and TFF3 knock-out mice had significantly smaller diameter of hepatic lobules compared to wild type mice. More connective tissue was found in TFF2 and TFF3 knock-out mice compared to wild type mice. TFF3 knock-out mice had more glycogen-positive cells than TFF2 knock-out mice and wild type mice, whose glycogen amounts did not differ statistically. CONCLUSION: There are significant histomorphological differences in liver structure of wild type mice and TFF2 and TFF3 knock-out mice in the diameter of hepatic lobules, amount of cell glycogen and connective tissue in the lobules.
