Objectives: To examine the distribution and number of interneurons in specific regions of genetically altered mice St3Gal2, St3Gal3 and St3Gal2/3 using specific antibodies (SST, VIP and nNOS). To examine whether there are differences in the distribution and number of interneurons between genetically modified mice and wild-type (WT) mice, and to examine the association of sialotransferase gene silencing with the distribution and ratio of interneurons in genetically modified mouse models. Study Design: An experimental case-control study. Material and methods: Brain tissue samples from genetically altered mouse models St3Gal2, St3gal3, St3Gal2/3 with a deficiency in the synthesis of complex gangliosides and a control wild-type mouse (WT) were used in the research. Immunohistochemical analysis was performed using SST, VIP, and nNOS antibodies. Photos were analyzed in the Fiji program, and the statistics were made in the program TIBCO Statistica ® 14.1.0 Results: By analyzing the distribution of SST, VIP and nNOS interneuron markers in the brains of genetically modified mice St3Gal2, St3Gal3 and St3Gal2/3, the distribution of interneurons was observed in all four primary regions of the neocortex M1, S1, V1, Au1. By comparing genetically modified mouse models with WT, changes in the number and distribution of interneurons were observed, however, the changes in the number of positive interneurons were not so large as to be statistically significant. Conclusion: The distribution of interneuron markers SST, VIP and nNOS was observed in all four primary regions of the neocortex. Comparison with WT did not demonstrate statistical significance.
