Abstract
Uvod: Rak vrata maternice u svijetu je drugi najčešći oblik raka u žena. Infekcija humanim papiloma virusom (HPV) najčešći je uzrok nastanka tog karcinoma. Istraživanja su pokazala da brojni čimbenici značajno povećavaju rizik za karcinom vrata maternice. Za onkogenezu HPV-a bitan je imunološki odgovor zaražene osobe. Dosadašnja istraživanja pokazala su da međudjelovanje HPV virusa i IL10 može dovesti do nastanka imunosupresivnog mikrookoliša u cerviksu, dok se prisutnost T alela IL-1B povezuje s kroničnom upalom i perzistentnom infekcijom HPV16/18. Ciljevi istraživanja: Analiza polimorfizama gena rs16944 (-511C/T) IL-1B i rs1800896 (-1082 A > G) IL-10 između ispitanica s dokazanim karcinomom vrata maternice i ispitanica u kontrolnoj skupini. Utvrditi postoji li statistički značajno odstupanje u pojavnosti polimorfizama gena IL-1B i IL-10 između ispitanica s dokazanim karcinomom vrata maternice i ispitanica u kontrolnoj skupini. Ispitanici i metode: Navedeni polimorfizmi gena analizirani su kod 81 ispitanice s dijagnozom vrata maternice i 80 ispitanica u kontrolnoj skupini. DNA je izolirana iz uzoraka pune krvi, a genotipizacija je učinjena metodom lančane reakcije polimerazom u stvarnom vremenu uz primjenu Taqman metodologije. Napravljena je statistička obrada dobivenih polimorfizama gena rs16944 (-511C/T) IL-1B i rs1800896 (-1082 A > G) IL-10. Rezultati: Učestalost rjeđeg alela G IL-10 rs1800896 (-1082 A > G) bila je 0,43 u kontrolnoj skupini, dok je kod ispitanica s dijagnozom vrata maternice iznosila 0,39. Učestalost rjeđeg alela T IL-1B rs16944 (-511C/T) u kontrolnoj skupini iznosila je 0,39 te 0,3 kod ispitanica s dijagnozom vrata maternice. Zaključak: Nije pronađena statistički značajna razlika u raspodjeli i učestalosti pojedinih alela i genotipova između ispitanica s dokazanim karcinom vrata maternice i kontrolne skupine.
Abstract (english)
Introduction: Cervical cancer is the second most common cancer in women worldwide. The most common cause of this cancer is the Human papillomavirus (HPV) infection. Research has shown that numerous factors significantly increase the risk of cervical cancer. An immune response is essential for the oncogenesis of HPV infected persons. To date, studies have shown that the interaction of the HPV virus and IL-10 can lead to the formation of an immunosuppressive microenvironment in the cervix. The presence of the T allele of IL-1B is associated with chronic inflammation and persistent infection of HPV16/18. Research objectives: Analysis of rs16944 (-511C/T) IL-1B and rs1800896 (-1082 A > G) IL-10 gene polymorphisms between subjects diagnosed with cervical cancer and subjects in the control group. The study aimed to determine whether there is a statistically significant difference in the incidence of IL-1B and IL-10 gene polymorphisms between subjects diagnosed with cervical cancer and subjects in the control group. Subjects and Methods: Pre-mentioned gene polymorphisms were analyzed in 81 subjects diagnosed with cervical cancer and 80 subjects in the control group. DNA was extracted from the peripheral blood, and genotyping was performed by a real-time polymerase chain reaction. Results: The frequency of the minor allele G IL-10 rs1800896 (-1082 A > G) was 0.43 in the control group, while in subjects diagnosed with cervical cancer was 0.39. The frequency of the minor allele T IL-1B rs16944 (-511C / T) was 0.39 in the control group, and 0.3 in patients diagnosed with cervical cancer. Conclusion: No statistically significant difference was found in the distribution and frequency of individual alleles and genotypes between subjects with proven cervical cancer and a control group.
