Abstract
Uvod: Kumarin ili 2H-kromen-2-on je heterociklički aromatski kemijski spoj koji je široko rasprostranjen u prirodi. Maligne bolesti predstavljaju jedan od najvećih problema u današnjoj medicini, a kumarini nalaze ulogu u liječenju istih ili nuspojava uzrokovanih nekom drugom vrstom terapije malignih bolesti. Cilj: Ispitati utjecaj derivata kumarina na aktivaciju apoptoze i autofagije u tumorskim stanicama in vitro. Materijali i metode: Proapoptotički učinak derivata kumarina (DK1, DK2 i DK3), u koncentraciji od 1x10-5 M, ispitan je na HeLa i MCF-7 staničnoj liniji, a određen protočnim citometrom. Rezultati: Svi testirani derivati djeluju na aktivaciju apoptoze u HeLa stanicama dok na MCF-7 stanice značajno utječe samo derivat DK3. Utjecaj na autofagiju je snižen u odnosu na kontrolu u obje stanične linije. Zaključak: Testirani derivati pokazuju povećan apoptotički učinak na HeLa staničnu liniju. U obje stanične linije ne potiču autofagiju.
Abstract (english)
Introduction: Coumarin, or 2H-chromen-2-one, is a heterocyclic aromatic chemical compound that is widespread in nature. Malignancies are one of the biggest problems in today's medicine and coumarins find a role in treating the same or side effects caused by some other type of malignancy therapy. Objective: To examine the influence of coumarin derivatives on the activation of apoptosis and autophagy in tumor cells in vitro. Materials and methods: The proapoptotic effect of coumarin derivatives (DK 1, DK 2 and DK 3), in concentration of 1x10-5 M, was examined on a HeLa and MCF-7 cell line, determined by a flow cytometer. Results: All tested derivatives affect the activation of apoptosis in HeLa cells, while MCF-7 cells are significantly affected only by the DK 3 derivative. The effect on autophagy was reduced in regard to both cell lines. Conclusion: The derivatives tested show an increased apoptotic effect on the HeLa cell line. In both cell lines they do not promote autophagy.
