Introduction: Psoriasis is a chronic inflammatory skin disease and one of the most common autoimmune diseases. It is associated with a higher risk of developing other diseases and has a significant impact on the quality of life. The development of psoriasis is influenced by a number of genetic and environmental factors with a disturbed skin barrier and the appearance of autoreactive immune cells. Results of many studies, which were mainly conducted on animal models, indicate a potential key role of γδ T cells in the progression and maintenance of psoriatic plaques, but their role and origin in psoriatic patients are not well understood. Objectives: Analyze and compare frequencies of peripheral CD3+TCRγδ+ T lymphocytes in the blood of healthy controls and subjects with psoriasis and examine their relation with disease severity and demographic characteristics of subjects. Participants and methods: Total peripheral mononuclear cells were isolated from collected peripheral blood of 22 healthy controls and 21 patients with psoriasis vulgaris. Phenotyping of γδ T lymphocytes was performed in a flow cytometer using FITC CD3 and PE-CY7 TCRγδ anti-human murine monoclonal antibodies. Results: There was no statistical difference in the relative frequencies of CD3+ γδ T lymphocytes between the healthy controls and psoriasis patients, most probably as a consequence of insufficiently specified subtypes of γδ T lymphocytes. A significant difference was observed in CD3-γδTCR+ cells that are not yet well understood. A significant correlation of CD3+γδTCRhigh T lymphocyte population with CMV IgG antibody levels was observed. Significant correlation of PASI and DLQI was found. Conclusion: In the examined sample the frequencies of CD3+ γδ T lymphocytes did not differ significantly between groups. In psoriatic patients CD3+γδTCRhigh cell frequency correlates with CMV IgG antibody levels.
