Introduction: Psoriasis vulgaris is a chronic autoimmune skin disease mediated by T lymphocytes. γδ T cells mediate important inflammatory effects in the affected parts of skin, mostly by adjusting the function and migration patterns at the blood – skin interface. Small noncoding microRNA molecules (hsa-microRNA-let7c, hsa-microRNA-20a, hsa-microRNA-29a, hsa-microRNA-92b) are key regulators of these cellular processes, but their role in peripheral γδ T lymphocytes of psoriasis patients is completely unexplored. Aim of the research: To examine transcriptional profiles of hsa-microRNA-let7c, hsamicroRNA-20a, hsa-microRNA-29a te hsa-microRNA-92b in γδT cells of peripheral blood of patients with psoriasis vulgaris and to establish their association with clinical, biochemical and anthropometric characteristics of psoriasis vulgaris. Material and method: The study included archival samples of RNA isolated from peripheral sorted γδ T lymphocytes of 16 healthy controls and 16 subjects suffering from psoriasis vulgaris. cDNA synthesis was performed using „TaqMan advanced cDNA synthesis kit“. Quantitative analysis of microRNA expression was performed on a QuantStudio 5 device by real-time polymerase chain reaction (qRT-PCR) using TaqMan Advanced microRNA probes and primere. Results: Compared to healthy subjects, peripheral γδ T lymphocytes of patients with psoriasis vulgaris show significantly reduced levels of microRNA hsa-20a-5p, while the expression of hsa-microRNA-92b-5p was significantly elevated. Loss of hsa-microRNA-20a-5p expression was associated with an increase in the peripheral share of Vδ1-Vδ2- subpopulation within the γδTCRint cell population and the expression profile of hsa-microRNA-20a-5p was positively correlated with ZBTB16 and the IL-18R gene expression. Conclusion: Psoriasis patients with milder clinical symptoms show changes in the expression of certain microRNAs, potentially important in the regulation of peripheral γδ T lymphocytes.
