Introduction: The increase of the amount of iron in neurons is connected to oxidative damage, amyloid accumulation, and neurodegeneration. Iron metabolism disorder occurs in Alzheimer's disease which is associated with chronic stress. Objectives: To construct immunochemical and histological coloring on iron and amyloid from brain tissue of young and old Sprague Dawleye males in order to determine the possible association of chronic stress, iron metabolism disorders, and neurodegeneration. Materials and Methods: The study included young and old Sprague Dawley male rats exposed to real or fake stress. The study included 40 Sprague Dawley male rats (4 groups, 10 animals per group) that were exposed to fake or real stress at 3.5 months or 14.5 months. The selected stressors were: nighttime alarm sounds, forced swimming in cold water, staying in a cold room, and exposure to light during the night. Animals weresacrificed 15 days after the last stress cycle. The sample archive was made of collected samples that were stored at -80 ° C or in the form of fixed tissue prepared for histological analysis. Results: Most iron and transferrin was found in the supstancia nigra(SN) cells of old animals exposed to sham stress, while the lowest values occurred in young sham animals. The most pTAUs were found in old animals exposed to chronic stress, and the least in young. Young animals accumulate more amyloid than the older ones. Conclusion: Aging causes iron accumulation in neurons of SN. Chronic stress causes the accumulation of iron in neurons of SN in the young animal group. The expression of transferrin reflects the amount of accumulated iron.
