Objectives: The aim of this study is to determine the percentage of cell survival in hepatic cell line (Huh7) model, after exposing the cells to different concentrations of amiodarone and tamoxifen in different periods of time. Another aim of the study is to determine the effect of different concentrations of amiodarone and tamoxifen on lipid droplet formation and distribution, as a marker of vesicular steatosis in hepatic cell line (Huh7) model. Study design: The study was conducted as a randomised controlled trial. Materials and methods: Hepatic (Huh7) cells were incubated in Dulbecco's Modified Eagle's medium (DMEM) supplemented with 10% Fetal Bovine Serum (FBS) and 1% Antibiotic/Antimycotic solution, in a humidified incubator supplied with 5% CO2 at 37 °C. After the cells had been grown in a six well plate, they were treated with 5µM and 10 µM of tamoxifen and amiodarone. Cell growth was monitored daily by light microscope and all the changes were shot with camera Olympus DP70, magnification 1:1000, after Oil Red O staining of the cells that were previously treated with 5µM and 10 µM of tamoxifen and amiodarone. In order to determine all the effects of amiodarone and tamoxifen on cell cultures, we used Trypan Blue staining, Oil Red O staining, DAPI staining and MTT analysis. Results: Treatment of hepatic (Huh7) cells with different concentrations of amiodarone and tamoxifen lead to massive increase in the number and size of lipid droplets in the cytosol of hepatic cells. The effect of different concentrations of amiodarone and tamoxifen lead to dose dependent decrease in the number of hepatic cells. Conclusion: Amiodarone and tamoxifen have an express lipogenic effect on triglyceride accumulation and steatogenesis in hepatocyte cell cultures. The steatogenic effect was characterised by lipid droplet formation.
