Abstract
Cilj: Imunohistokemijskim metodama, uz pomoć protutijela na inzulinski receptor alfa (IR-α) obojiti tkiva maternice štakorica hranjenih standardnom prehranom ili prehranom bogatom mastima i šećerima od 3. tjedna starosti te usporediti izražaj inzulinskih receptora u tkivu miometrija štakorica hranjenih standardnom prehranom i prehranom bogatom mastima i šećerima. Ustroj studije: Eksperimentalna studija na animalnom modelu – studija parova. Materijali i metode: Studija je napravljena na arhivi tkiva maternica prikupljenih u sklopu studije RECOOP-CMSC Senior Scientist (RCSS) Grant 2018-2020 #012: „The role of obesity-induced low-grade inflammation in the adipokine signaling in pregnant rat uterus“. U istraživanju su korištene ženke Sprague-Dawley štakora koje su od 3. tjedna starosti hranjene standardnom prehranom (SD) ili prehranom bogatom mastima i šećerima (HFHSD). Prisutnost IR-α u tkivu maternice analizirana je imunohistokemijskom metodom. Nakon fiksacije u 4 %-tnoj puferiranoj otopini formaldehida tijekom 24 sata, tkiva su uklopljena u parafinske blokove standardnim postupkom. Na deparafiniziranim rezovima debljine 5 µm provedeno je imunohistokemijsko bojanje tkiva uz pomoć protutijelo na IR-α. Na kraju je provedena analiza mikrofotografija te statistička analiza dobivenih podataka. Rezultati: U vanjskom miometriju štakorica hranjenih prehranom bogatom mastima i šećerima (HFHSD) dolazi do povećanja izražaja inzulinskih receptora alfa u odnosu na štakorice hranjene standardnom prehranom (SD). Zaključak: Imunohistokemijskom metodom uz pomoć protutijela na IR-α obojana su tkiva maternica te je zapaženo povećanje izražaja IR-α u vanjskom miometriju HFHSD skupine.
Abstract (english)
Objective: Immunohistochemical methods, with the help of antibodies to the insulin receptor alpha (IR-α), to stain the uterine tissues of rats fed a standard diet or a diet rich in fats and sugars from the 3rd week of age and to compare the expression of insulin receptors in the myometrial tissue of rats fed standard diet and diet rich in fats and sugars. Study desing: Experimental research on an animal model – the case-control study. Material and methods: The research was made on the archive of uterine tissues collected as part of the study RECOOP-CMSC Senior Scientist (RCSS) Grant 2018-2020 #012: „The role of obesity-induced low-grade inflammation in the adipokine signaling in pregnant rat uterus“. The research used female Sprague-Dawley rats that were fed a standard diet (SD) or a diet rich in fats and sugars (HFHSD) from the 3rd week of age. The presence of IR-α in uterine tissue was analyzed by immunohistochemical method. After fixation in 4 % buffered formaldehyde solution for 24 hours, the tissue were embedded in paraffin blocks using a standard procedure. Immunohistochemical tissue staining was performed on deparaffinizde sections with a thickness of 5 µm using an antibody to IR-α. At the end, an analysis of microphotographs and statistical analysis of the obtained data was carried out. Results: In the outer myometrium of rats fed a diet rich in fats and sugars (HFHSD), there is a increase in the expression of insulin receptors alpha compared to rats fed a standard diet (SD). Conclusion: Uterine tissues were stained with an immunohistochemical method using IR-α antibodies, and an increase expression was observed in the outer myometrium of the HFHSD group.
